If you are talking adwords then a really important thing to do is to list negative keywords - anything you can think of that searches for your main keywords would be looking for that you do not have, and therefor do not want to pay to get them to your site. An example would be an Apple Computer seller minusing "fruit".
What I thought this topic was about is what has been called the "Long Tail" - the vast niche markets that the Internet opens up. If you sell goldfish exercise equiptment there probably is not enough people in your area to support a business selling it. :'( But with the vast reach of the internet there may be enough people around the world interested in stronger goldfish where you could make a nice living. (Shipping costs would not be too much either)
